An enzyme-linked immunosorbent assay (ELISA) was developed for the quantitative determination of lipoxygenase-1 (L1) and lipoxygenase- 3 (L3) in soybeans (Glycine max (L.) Merr.). Monospecific goat (Carpa hircus) and rabbit (Oryctolagus cuniculus) immunoglobulins purified by affinity chromatography were used for the L1 and L3 assays, respectively. In both cases the coefficients of variation in the assays were about 10%. A linear relationship between lipoxygenase (L1) enzymic activity and the enzyme protein content as determined by ELISA was obtained. The technique has considerable potential for screening of segregating soybean populations for genotypes deficient in specific lipoxygenase isozymes.
